No, this is designed specifically for Gen 3 frames only. Gen 4 and Gen 5 Glocks have different frame dimensions and won't accept this magwell. Strike Industries makes separate magwell models for each generation, so make sure you're ordering the right one for your pistol.
Not with standard Glock backstraps. The magwell won't seat properly if a backstrap is installed on the frame. You'd need to remove it for the magwell to fit. If you run backstraps regularly, this might not be the best choice for your setup.
Hand-tight is plenty. Strike Industries specifically warns against overtightening the M2.5 set screw, as it can throw off the magwell's alignment and damage both the part and your frame. If it feels secure by hand, stop there. When in doubt, a light touch beats risking damage.
Most aftermarket base plates and extensions work fine, but compatibility isn't guaranteed with every brand. Some less common floor plates might have fitment issues.
